Promene u genomu limfocita periferne krvi pacijenata sa kardiovaskularnim bolestima: faktori rizika i protektivni efekat gljive Lenzites betulinus (L.) Fr.

Cilj disertacije je bio da se ispitaju promene u genomu pacijenata sa akutnimkoronarnim sindromom (AKS) i srčanom insuficijencijom (SI) u odnosu na faktorerizika procenjivane na nivou molekula DNK i hromozoma, efekat koronarneangiografije (KA) i potencijalni protektivni efekat gljive Lenzites betulinus uodnosu na njihov fitohemijski sastav. Nivo DNK oštećenja je analiziran komettestom, a frekvenca hromozomskih aberacija je analizirana citokinezis-blokmikronukleus testom. Genotoksični i genoprotektivni potencijal različitihekstrakata gljive je analiziran komet testom, a fitohemijski sastav je ispitivanspektrofotometrijskim i hromatografskim analizama. Rezultati su pokazali dapacijenti imaju značajno veći nivo genomskog oštećenja/genomske nestabilnosti uodnosu na zdrave osobe, kao i povećano oštećenje genoma nakon KA u poređenju saoštećenjem pre KA. Faktori rizika (pozitivna porodična anamneza, hipertenzija,hiperholesterolemija, terapija lekovima, srčani biomarkeri i doza zračenja pri KA)su uticali na promene u genomu pacijenata. Ekstrakti gljive su neznačajno povećavalinivo DNK oštećenja u limfocitima zdravih osoba, dok su značajno redukovaliterapijom-indukovan i zračenjem-indukovan nivo DNK oštećenja u limfocitimapacijenata. Gljiva je bila bogata mineralima, organskim i masnim kiselinama,šećerima i polifenolnim jedinjenjima. Može se zaključiti da su pacijenti sa AKS iSI imali značajno veće promene u genomu u odnosu na zdrave osobe, što je ukorelaciji sa ozbiljnošću bolesti i faktorima rizika. L. betulinus se može smatratigljivom visoke nutritivne i funkcionalne vrednosti, jer ekstrakti gljive nisu biligenotoksični u ispitivanim koncentracijama u kultivisanim limfocitima zdravihosoba, dok su kod pacijenata ispoljili hemo- i radioprotektivni efekat protivterapijom/zračenjem-indukovanog DNK oštećenja.

The dissertation aimed to examine the changes in the genome of patients with acutecoronary syndrome (ACS) and heart failure (HF) in relation to risk factors assessed at thelevel of DNA and chromosome, the effect of coronary angiography (CAG), and the potentialprotective effect of Lenzites betulinus mushroom in relation to their phytochemicalcomposition. The level of DNA damage was analyzed by the comet assay, and frequency ofchromosomal aberration was analyzed by the cytokinesis-block micronucleus test. Thegenotoxic and genoprotective potential of different mushroom extracts was assessed by thecomet assay, and their phytochemical composition was examined by spectrophotometric andchromatographic analyses. The results showed significantly higher level of genomedamage/instability in patients compared to healthy individuals, as well as increased genomedamage after CAG compared to damage before CAG. Risk factors (positive family history,hypertension, hypercholesterolemia, drug therapy, cardiac biomarkers and dose of Xradiation during CA) affected the level of genome damage in patients. Mushroom extractsinsignificantly increase the level of DNA damage in the lymphocytes of healthy individuals,but significantly reduced the therapy-induced and radiation-induced level of damage in thelymphocytes of patients. The mushroom was rich in minerals, sugars, organic and fatty acids,and polyphenolic compounds. It can be concluded that patients with ACS and HF hadsignificantly higher changes in the genome than healthy individuals, which is correlated withthe severity of the disease and risk factors. L. betulinus may be considered a mushroom ofhigh nutritional and functional value, as its extracts were non-genotoxic at testedconcentrations in cultured lymphocytes of healthy individuals and exhibited a chemo- andradioprotective effect against therapy/radiation-induced DNA damage in patients.
