Uloga ekspresije COPB2 i procesa apoptoze u nastanku karcinoma endometrijuma

U nastanku karcinoma endometrijuma važnu ulogu imaju geni zaduženi zasintezu proteina koji učestvuju u ćelijskim tarnsportnim i metaboličkim procesima.Proteini familije Bcl-2 ključni su unutarćelijski modulatori procesa apoptoze. Udo sada rađenim studijama dokazan je značaj COPB2 u nastanku i razvojuadenokarcinoma.Cilj ove studije je ispitati uticaj poremećaja apoptoze i ekspresije gena zaCOPB2 na nastanak karcinoma endometrijuma.Uzimalo se tkivo endometrijuma od pacijentkinja eksperimentalne (40ispitanica) i kontrolne grupe (20 ispitanica). Metodom reverzne transkripcijekvantitativnog PCR-a određivala se relativna ekspresija gena za COPB2 i proteinBcl-2 u endometrijalnim ćelijama. Ispitana je uloga poremećaja regulacije apoptozepreko Bcl-2, kao i ekspresije gena za COPB2 u nastanku karcinoma endometrijuma.Utvrđeno je da postoji statistički značajna razlika (R<0,005) u vrednostimaekspresije gena COPB2 i Bcl-2 između pacijentkinja sa i bez karcinomaendometrijuma. Za oba gena je prikazan značajan dijagnostički potencijal u razdvajanjuobolelih od zdravih pacijentkinja, s' tim što je dijagnostički potencijal COPB2 genaspecifičniji. Za Bcl-2 površina ispod krive (AUC) iznosi 0,666 i ovaj rezultat jestatistički značajan (r=0,038). Za COPB2 površina ispod krive (AUC) iznosi 0,877 iovaj rezultat je statistički značajan (r=0,000).Vrednosti relativne ekspresije COPB2 i Bcl-2 su snižene u početnimstadijumima karcinoma endometrijuma. Njihov poremećaj može imati značajnu ulogu unastanku karcinoma endometrijuma.

SUMMARY:Genes responsible for the synthesis of proteins involved in cellular transport andmetabolic processes play an important role in the development of endometrial carcinoma.Bcl-2 family proteins are key intracellular modulators of the apoptosis process. Previousstudies have demonstrated the importance of COPB2 in the development and progression ofadenocarcinoma.The aim of this study is to examine the impact of apoptosis disorders and COPB2 geneexpression on the development of endometrial carcinoma.Endometrial tissue was taken from patients in the experimental group (40 patients) andthe control group (20 patients). The relative expression of the COPB2 gene and Bcl-2 proteinin endometrial cells was determined using the reverse transcription quantitative PCR method.The role of apoptosis regulation disorders via Bcl-2, as well as COPB2 gene expression in thedevelopment of endometrial carcinoma, was examined.It was found that there is a statistically significant difference (p<0.005) in COPB2 andBcl-2 gene expression values between patients with and without endometrial cancer. For bothgenes, a significant diagnostic potential was shown in separating cancer patients from healthypatients, with the fact that the diagnostic potential of the COPB2 gene is more specific. ForBcl-2 area under the curve (AUC) is 0.666 and this result is statistically significant (p=0.038).For COPB2 the area under the curve (AUC) is 0.877 and this result is statistically significant(p=0.000).The relative expression values of COPB2 and Bcl-2 are decreased in the initial stagesof endometrial carcinoma. Their disorder can play a significant role in the development ofendometrial cancer.
