Efekti modulacije metabolizma kreatina na funkciju srca pacova izloženih intervalnom treningu visokog inteziteta

Uvod: Poslednjih godina, intervalni trening visokog intenziteta u kombinaciji sarazličitim ergogenim sredstvima postao je veoma popularan modalitet treninga međusportistima.Cilj: Cilj ove studije je da se ispitaju efekti samostalne i udružene suplementacijekreatinom, betainom, gvanidinoacetatom kod treniranih pacova na funkciju srca,oksidacioni stres i na morfološku građu srca.Metode: U studiju je uključeno 72 pacova (Wistar albino soja, muškog pola, starosti 8nedelja) razvrstani u 9 grupa: 1. KON- kontrola, 2. T- trening, 3. T+B – trening i betain,4. T+K – trening i kreatin, 5. T+B+K – trening i betain i kreatin 6. T+G– trening igvanidinoacetat, 7. T+G+K – trening i gvanidinoacetat i kreatin, 8. T+G+B – treningi gvanidinoacetat i betain, i 9. T+G+K+B – trening i gvanidinoacetat, kreatin ibetain. Izolovano srce pacova je perfundovano metodom retrogradne perfuzije poLangendorff-u i praćeni su sledeći parametri: dp/dt max, dp/dt min, SLVP, DLVP i HR.U uzorcima koronarnog venskog efluenta i u krvi pacova određivani su parametrioksidacionog stresa. Izolovana srca, kvadriceps, jetra i bubrezi su ritinskipripremljeni za histološku analizu.Rezultati:. Suplementacija kreatinom, betainom i gvanidinoacetatom poboljšavafunkciju miokarda, snižava vrednosti prooksidanasa a povećava vrednostiantioksidanasa. Najveći stepen hipertrofije kardiomicita, najveće povećanje depoaSERCA pumpe u srcu, kao i najveće smanjenje sadržaja kolagena u srcu bilo je u grupiživotinja koje su trenirale i bile na suplementaciji gvanidinoacetatom.Zaključak: Suplementacija erogenim sredstvima kod treniranih pacova poboljšavakardiodinamske parametre i oksidacioni stres izolovanog srca pacova.

Introduction: In recent years, high-intensity interval training combined with variousergogenic means has become a very popular training modality among athletes.Objective: The aim of the study is to examine the effects of independent and combinedsupplementation with creatine, betaine, guanidinoacetate in trained rats on cardiac function,oxidative stress and morphological structure of the heart.Methods: The study included 72 rats (Wistar albino, male, 8 weeks old) divided into 9 groups:1. KON- control, 2. T- training, 3. T+B- training and betaine, 4. T+K– training and creatine, 5.T+B+K– training and betaine and creatine 6. T+G– training and guanidinoacetate, 7. T+G+K–training and guanidinoacetate and creatine, 8. T+G+B– training and guanidinoacetate andbetaine, and 9. T+G+K+B– training and guanidinoacetate, creatine and betaine. The isolatedrat heart was perfused using the Langendorff retrograde perfusion method and the followingparameters were monitored: dp/dt max, dp/dt min, SLVP, DLVP and HR. Oxidative stressparameters were determined in the samples of coronary venous effluent and in the blood ofrats. Isolated hearts, quadriceps, livers and kidneys were routinely prepared for histologicalanalysis.Results: Supplementation with creatine, betaine and guanidinoacetate improves myocardialfunction, lowers prooxidant values and increases antioxidant values. The highest degree ofcardiomyocyte hypertrophy, the highest increase in the SERCA pump depot in the heart, aswell as the highest decrease in collagen content in the heart was in the group of animals thattrained and were supplemented with guanidinoacetate.Conclusion: Supplementation with erogenous agents in trained rats improves cardiodynamicparameters and oxidative stress in the isolated rat heart.
