Uticaj kombinovanog tretmana alogenim matičnim ćelijama iz masnog tkiva i hiperbaričnom oksigenacijom na progresiju eksperimentalnog gonartritisa kod pacova

Blagotvorno dejstvo hiperbarične oksigenacije (HBO) u inflamatornim procesimačini ga atraktivnom vrstom lečenja hroničnog artritisa. Efekti kombinovane terapije zasnovane na primeni alogenih matičnih ćelija masnog tkiva (ADS) i HBOna progresiju osteoartritisa (OA) nisu u potpunosti istraženi.Aktuelna studija je istraživala efikasnost intraartikularne primene ADS u kombinaciji sa tretmanom HBO u modelu OA pacova. Model OA kod pacova indukovan je intraartikularnom injekcijom monojodoacetata(MIA) i 7 dana nakon primene MIA pacovi su podeljeni u osam grupa: zdrava kontrola(CTRL), OA, ADS, HBO, ADS+HBO21day, ADS+HBO28day, 0,9% NaCl, medijum zauzgajanje ćelija (MED). 1h106 ADS ubrizgana je u zglob kolena samostalno ili u kombinaciji sa HBO tretmanom. Rezultati lečenja su procenjeni radiografskom, histološkom analizom, specifičnim bojenjem zglobne hrskavice, određivanjeminflamatornih i pro/antioksidativnih markera.Kombinovani tretman značajno poboljšava regeneraciju hrskavice u kolenskom zglobu. Rtg skor oštećenja kolenskog zgloba je značajno smanjen u grupamaADS+HBO21day i ADS+HBO28day u poređenju sa OA. Međutim, pozitivan efekat uADS+HBO28day grupi bio je veći nego u ADS+HBO21day grupi. Zglobna hrskavica je bila relativno normalna u grupi ADS+HBO28day, ali umerena degeneracija je primećena u ADS+HBO21day u poređenju sa OA grupom. Ovi nalazi su u skladu sahistopatološkim rezultatima. Znatno niži nivo O2−. primećeno je u grupi ADS+HBO28day, ali i viši nivo NO u poređenju sa ADS+HBO21day grupom. Štaviše, u grupi ADS+HBO28day primećene su značajno veće koncentracije IL-10, ali nije bilo značajne razlike u proinflamatornim citokinima u uzorcima seruma.Ovi rezultati ukazuju da je jedna intraartikularna injekcija ADS u kombinaciji saHBO efikasno ublažila progresiju OA.

Hyperbaric oxygenation therapy (HBO) is a promising kind of treatment for chronic rheumatoid arthritis because of its positive effects on inflammatory processes. Furthermore, the effects of combination therapy based on adipose stem cells (ADS) and HBO on the development of osteoarthritis (OA) have not been thoroughly studied.The goal of the current investigation was to determine if intra-articular injections of ADS and HBO were effective in treating a rat OA model.The OA rat model was induced by intra-articular injection of monoiodoacetate (MIA), and 7 days after MIA application, rats were divided into eight groups: healthy (CTRL), OA, ADS, HBO, ADS+HBO21day, ADS+HBO28day, 0,9% NaCl, medium (MED). In the knee joint, alone or in combination with HBO, 1x106 ADS was administered. Results of treatment were assessed using articular cartilage specific staining, histological and radiographic analysis and determining of inflammatory and pro/antioxidative markersADS and HBO together promoted cartilage regeneration in the knee joint. In comparison to the OA, the Rtg score for knee joint damage was significantly lower in the HBO+ADS21day and HBO+ADS28day groups. The HBO+ADS28day group experienced a bigger favorable impact than the HBO+ADS21day group. Cartilage in the HBO+ADS28day group was normalbut moderate degeneration was observed in ADS+HBO21day compared to OA group. These results concur with the histological findings. HBO+ADS28day group had a lower O2level than HBO+ADS21day group, but a higher NO level and greater IL-10 concentrations, although there was no discernible difference in proinflammatory cytokine levels in serum samples.An intra-articular injection of ADS and HBO effectively slowed the progression of OA.
