Sinteza, karakterizacija i biološka aktivnost kompleksa Pd(II) sa etilendiaminskim i kumarinskim derivatima aminokiselina

U ovoj Doktorskoj disertaciji opisana je sinteza i karakterizacija nekoliko novih liganada, etilendiaminskih i kumarinskih derivata nekih aminokiselina i odgovarajućih paladijum(II)-kompleksa. Najpre je opisana sinteza i karakterizacija O,O'-dialkil estara (S,S)-etilen--diamin-N,N'-di-(2,2'-di-(4-hidroksi-benzil))sirćetne kiseline (H2-(S,S)-eddtyr). Ova jedinjenja su dobijena direktnom reakcijom između tetradentatnog liganda (S,S)-etilendiamin-N,N'-di-(2,2'-di-(4-hidroksi-benzil))sirćetne kiseline i odgovara- -jućeg apsolutnog alkohola (etanol, 1-propanol, 1-butanol i 1-pentanol) u molskom odnosu 1 : 2, uz uvođenje gasovitog hlorovodonika. Takođe, prikazana je i sinteza kumarinskih derivata nekih aminokiselina. Pomenuti ligandi dobijeni su refluktovanjem smeše 3-acetil-4-hidroksi kumarina, hidrohlorida metil estara aminokiselina S-valina, glicina, S-metionina ili S-triptofana i trimetilamina. Tok reakcije je praćen tankoslojnom hromatografijom (toluen : aceton = 8 : 2). Nakon završene reakcije, uparavanjem rastvarača do polovine svoje zapremine i dodavanjem 5 cm3 vode, dobijeni sukumarinski derivati sa pomenutim aminokiselinama. Strukture i sastav izolovanih liganada pretpostavljene su primenom elementalne mikroanalize, infracrvene, 1H i 13C NMR spektroskopije, a potvrđene na bazi rezultata rendgenske strukturne analize u slučaju metil 2-[(1-(2,4-dioksohroman-3-iliden)etilamino-3-metil)]butanoata. Dobijeni O,O'-dialkil estri (S,S)-etilendiamin-N,N'-di-(2,2'-di-(4-hidroksi--benzil))sirćetne kiseline su upotrebljeni za sintezu odgovarajućih paladijum(II)--kompleksa. Sastav dobijenih kvadratno-planarnih paladijuma(II)-kompleksa potvrđen je elementalnom analizom, dok je struktura izolovanih kompleksa pretpostavljena na osnovu infracrvene, 1H i 13C NMR spektroskopije.
The synthesis and characterization of several new ligands, ethylenediamine and coumarin derivatives of some amino acids and corresponding palladium(II)-complexes are described in this PhD thesis. Firstly, the synthesis and characterizations of O,O'-dialkyl esters of (S,S)-ethylenediamine- -N,N'-di-(2,2'-di-(4-hydroxy-benzyl)acetic acid, H2-(S,S)-eddtyr are described. These compounds were obtained by direct reaction between tetradentate ligand (S,S)-ethylenediamine-N,N'-di-(2,2'- -di-(4-hydroxy-benzyl)acetic acid and the corresponding absolute alcohol (ethanol, 1-propanol, 1-butanol, 1-pentanol) in a molar ratio 1:2 with the addition of gaseous hydrogen-chloride. Also, the synthesis of coumarin derivatives of some amino acids are also shown. Mentioned ligands were obtained refluxing a mixture of 3-acetyl-4-hydroxy coumarin, hydrochloride methyl esters of amino acids S-valine, glycine, S-methionine or S-tryptophan and trimethylamine. The progress of the reactions was monitored by TLC (toluene : acetone = 8 : 2). After the finishing of the reactions, the evaporation of solvent to the half of native volume and addition of 5 cm3 of water, the obtained coumarin derivatives with mentioned amino acids were obtained. The obtained O,O'-dialkyl esters of (S,S)-ethylenediamine-N,N'-di-(2,2'-di-(4-hydroxy- -benzyl)acetic acids are used for synthesis of the corresponding palladium(II)-complexes. The composition of obtained square-planar palladium(II)-complexes was confirmed by elemental microanalysis, and the structure of isolated complexes is assumed on basis of infrared, 1H and 13C NMR spectroscopy.
