Аntihypertensive drugs have positive benefits in cardiovascular disease, so we sought toexamine their effect on ischaemia-reperfusion (I/R) injury of heart.The study's objective was to ascertain results of subchronic therapy of аntihypertensive agentson I/R dysfunction of the heart in spontaneously hypertensive rats (SHR) by a model of globalischemia on isolated heart of rat.The research included 80 Wistar Kyoto SHR, male sex, grouped into 8 groups. The animalswere treated 4 weeks (10 mg/kg) with hydrochlorothiazide, indapamide, spironolactone,zofenopril or their combinations (ZHS, ZIS) per os. After treatment we analysed hemodynamicmeasurements, echocardiography, and myocardial function according to the Langendorffretrograde perfusion method. An induced global ischemia model involved applying 20 minutesof ischemia followed by 30 minutes of reperfusion to the heart. Oxidative stress markers weredetermined spectrophotometrically from coronary venous effluent, plasma and erythrocytelysate. Heart and kidney tissue samples were pathohistologically analyzed.Treatment with ZHS and ZIS lowered the blood pressure of SHR, alleviated left ventricularhypertrophy and increased the ejection fraction, while it had no effect on the oxidation status.On the other hand, all three tested diuretics showed exceptional antioxidant potential.Pathohistological analysis indicates that subchronic administration of antihypertensive agentsdoes not lead to significant changes in heart and kidney. Cardiac function monitoring of SHRprovided insight into cardioprotective effects of indapamide.

Kako su višedecenijska istraživanja pokazala kardioprotektivna dejstvaantihipertenzivnih lekova, zainteresovalo nas je da istražimo njihov efekat naishemijsko-reperfuzionu (I/R) povredu srca.Cilj studije je bio da se utvrde efekti subhronične terapije različitimantihipertenzivnim lekovima na I/R disfunkciju srca kod hipertenzivnih pacova pomodelu globalne ishemije na izolovanom srcu pacova.Istraživanje je sprovedeno na 80 spontano hipertenzivnih Wistar Kyoto pacovamužjaka, grupisanih u 8 grupa. Životinje su tretirane četiri nedelje sa 10 mg/kghidrohlorotiazida, indapamida, zofenoprila, spironolaktona ili njihovihkombinacija (ZHS, ZIS) per os. Nakon tretmana sprovedena su hemodinamska merenja,ehokardiografija i procena funkcije miokarda prema Langendorff retrogradnoj metodiperfuzije. Indukovani model globalne ishemije je podrazumevao primenu 20 minutaishemije, a zatim 30 minuta reperfuzije na srce (I20:R30). Markeri oksidativnogstresa određivani su iz koronarnog venskog efluenta, plazme i lizata eritrocitaspektrofotometrijski. Uzorci tkiva srca i bubrega su patohistološki analizirani.Tretman kombinacijom ACE-inhibitora i duretika značajno je snizio krvni pritisakspontanohipertenzivnih pacova, ublažio hipertrofiju leve komore i povećaoejekcionu frakciju, dok na oksidacioni status nije delovao. S druge strane samostalnaprimene sva tri ispitivana diuretika je pokazala izuzetan antioksidativnipotencijal. Patohistološka analiza uzoraka tkiva srca i bubrega, ukazuje dasubhronična primena antihipertenzivnih agenasa ne dovodi do značajnih promena uovim organima. Praćenje srčane funkcije spontanohipertenzivnih pacova ex vivo daloje uvid u kardioprotektivne efekte indapamida.
